PURPOSES OF INVESTIGATION Ordinarily complement is titrated by mixing the complement serum, hemolysin and corpuscles at once and incubating at 38 C. for 'Investigation aided by funds accruing from the preparation of arsphcnamine. one hour; as shown in a previous article (2) some destruction of complement inevitably occurs during a primary incubation which is particularly marked when the complement is titrated in the presence of antigen with a primary incubation of four to eighteen hours at 8 C. to 10 C. with or without an additional incubation at 38 C. For this reason it is important to study the influence of temperature and duration of primary incubation upon nonspecific complement fixation or anticomplementary activity by antigen alone and serum alone, which constitute the purposes of this investigation in relation to studies bearing upon the primary incubation for complement-fixation tests.
